2-up keeps text comfortably readable and is the safe choice for documents you will actually read. 4-up suits slide handouts, since slides carry large type. 9-up and 16-up are really for overviews — seeing the shape of a document at a glance rather than reading it.
That depends on the original type size and how many pages you fit. Halving the linear scale at 4-up turns 11pt body text into roughly 5.5pt, which is hard going in print. Slides and large-format pages survive far better than dense reports, so check one sheet before printing fifty.
Handouts are the common case — giving an audience slides they can annotate without one slide per sheet. It is also good for proofing a layout, since seeing sixteen pages together reveals inconsistent headings, spacing and structure that you would never notice scrolling one page at a time.
